摘要:建立高效阴离子交换色谱-脉冲安培检测法测定生态养护抑菌凝胶中褐藻寡糖含量的方法。采用阴离子交换柱CarboPac PA10(250 mm×2 mm,3.5 μm),以氢氧化钠、乙酸钠和水为洗脱液进行梯度洗脱,柱温为35℃,流量为0.8 mL/min。α-L-古罗糖醛酸单糖(PG)和β-D-甘露糖醛酸单糖(PM)的质量浓度分别在0.25~9.0 μg/mL和0.18~8.4 μg/mL范围内与色谱峰面积线性关系良好,线性相关系数分别为0.999 7和0.999 8,检出限分别为0.06, 0.03μg/mL,定量限分别为0.20,0.10μg/mL。测定结果的相对标准偏差分别为1.11%,1.25% (n=6),加标回收率分别为101.03%和95.22%。该方法简便易行,灵敏度高,重现性好,适用于生态养护抑菌凝胶中褐藻寡糖的分析。 
Abstract:The method for the determination of AOS in ecological conservation serum gel was established by high performance anion exchange chromatography with pulsed ampere detection (HPAE-PAD). The analysis was performed on a CarboPac PA10 column (250 mm×2 mm, 3.5 μm). With the gradient elution using NaOH, NaAc and H2O as the mobile phase. The column temperature was 35℃, and the flow rate was 0.8 mL/min. The mass concentration of α-L-guluronic acid monosaccharide (PG) and β-D-mannuronic acid monosaccharide (PM) had a good linear relationship with peak area in the range of 0.25-9.0 μg/mL and 0.18-8.4 μg/mL, respectively. The linear correlation coefficient was 0.999 7 and 0.999 8, the detection limit was 0.06, 0.03 μg/mL, the quantitative limit was 0.20, 0.10 μg/mL. The relative standard deriation of detection results of PG and PM was 1.11% and 1.25% (n=6), the recovery rate was 101.03% and 95.22%, respectively. The method was simple, sensitive and reproducible which is suitable for the analysis of alginate oligosaccharides in the ecological curing gel.
