摘要:建立高效液相色谱法测定白花蛇舌草中对香豆酸和芦丁含量的方法。色谱柱:Kromasil C18(250 mm×4.6 mm,5 μm);流动相:甲醇-0.1%磷酸溶液(30∶70);流量:1.0 mL/min;对香豆酸和芦丁的检测波长分别为308 nm和353 nm;柱温:30 ℃。对香豆酸的质量浓度在2.11~12.66 最/mL范围内与其色谱峰面积具有良好的线性关系(r=1.0000);芦丁的质量浓度在8.34~50.02 最/mL范围内与其色谱峰面积具有良好的线性关系(r=1.0000)。对香豆酸、芦丁的平均回收率分别为94.01%、91.05%。对香豆酸、芦丁6次测定结果的相对标准偏差分别为1.60%、1.16%。该方法可用于同时测定白花蛇舌草中对香豆酸和芦丁含量。
Abstract:The method for determination of p-Coumaric acid and Rutin in Oldenlandia Diffusa(Willd.) Roxb. by HPLC was established. The chromatographic column was Kromasil C18 (250 mm×4.6 mm, 5 洀) and the column temperature was 30℃.
The mobile phase was methanol-0.1% H3PO4 water solution(volume ratio was 30∶70) with the flow rate of 1.0 mL/min. The detection wavelengths of p-Coumaric acid and Rutin were 308 nm and 353 nm, respectively. The mas concentration of p-Coumaric acid had good linear relationship with the chromatographic peak area in the range of 2.11-12.66
最/mL with r=1.0000. The mas concentration of Rutin had good linear relationship with the chromatographic peak area in the range of 8.34-50.02 最/mL with r=1.0000. The average recovery of p-Coumaric acid and Rutin were 94.01% and 91.05%, respectively. The relative standard deviations of determination results for p-Coumaric acid and Rutin were 1.60%, 1.16%(n=6), respectively. The method is suiatble for the simultaneous determination of p-Coumaric acid and Rutin in Oldenlandia diffusa(Willd.) Roxb.
