摘要: 基于特征图谱并根据主要成分含量比例特征,建立一种鉴别制草乌及其伪品的方法。采用 Waters XBridge C18 (250 mm×4.6 mm,5 μm) 色谱柱,以乙腈 -四氢呋喃(体积比为 25∶15)和 0.1 mol/L 乙酸铵水溶液为 流动相,检测波长为 235 nm,流量为 1.0 mL/min,柱温为 30 ℃,用高效液相色谱进行测定生物碱;采用 ChemPattern 化学计量学软件对采集的数据进行处理分析。制草乌和伪品中 3 种成分的含量比例显著不同,制草乌中苯甲酰新乌 头原碱为含量最高的成分,伪品中苯甲酰乌头原碱含量最高。苯甲酰新乌头原碱与苯甲酰乌头原碱含量比值在正、 伪品中差异大,正品约为 10,伪品约为 3/100 ;苯甲酰次乌头原碱与苯甲酰乌头原碱含量比值差异大,正品约为 1, 伪品约为 3/100。该法建立的制草乌特征图谱专属性强,可结合 3 种主要成分含量的比例特征鉴别正、伪品,为规范 制草乌药材的使用提供科学依据。  
Abstract:A method for identifying Radic aconiti kusnezoffii preparata and its counterfeits was established based on characteristic atlas and content proportion characteristics of main components. Waters XBridge C18 column (250 mm×4.6 mm, 5 μm) was used, the mobile phase was acetonitrile-tetrahydrofuran and 0.1 mol/L ammonium acetate solution, the flow rate was 1.0 mL/min, and the detection wavelength was 235 nm, as well as the column temperature was 30 ℃. Determination of alkaloids was realized by high performance liquid chromatography (HPLC). Chemometrics software ChemPattern was employed to analyze the research data. The content ratio of the three components in Radic aconiti kusnezoffii preparata and the fake was significantly different. The content of benzoyl neoaconitine in Radic aconiti kusnezoffii preparata was the highest, and the content of benzoyl aconitine in the fake was the highest. The content ratio between benzoylmesaconine and benzoylaconitine was 10 in authentic herb, 1/100 in the counterfeit herb. The content ratio between benzoylmesaconine and benzoylaconitine was 1 in authentic herb, 3/100 in the counterfeit herb. The characteristic atlas of Radic aconiti kusnezoffii preparata established by this method is highly exclusive, and it can be combined with the proportional characteristics of the contents of three main components to identify the true and false products, so as to provide a scientific basis for standardizing the use of Radic aconiti kusnezoffii preparata.
