摘要: 建立超高效液相色谱 -串联质谱法同时测定马铃薯中缬氨酸、苏氨酸、亮氨酸、异亮氨酸、赖氨酸、甲硫氨 酸、苯丙氨酸、色氨酸8种必需氨基酸的分析方法。样品经超纯水提取,用Accucore HILIC 色谱柱进行分离,以含0.1% (体积分数)甲酸的甲醇溶液为流动相进行洗脱,流量为 0.2 mL/min,采用电喷雾电离源,正离子扫描,多反应监测模 式进行检测。8 种氨基酸在各自质量浓度范围内与色谱峰面积呈线性关系良好,线性相关系数为 0.999 0~0.999 6,检 出限为 0.04~1.50 mg/kg,定量限为 0.12~4.99 mg/kg。测定结果的相对标准偏差为 0.4%~4.6%(n=6),且在不同添 加水平下的样品回收率为 89.9%~115.1%。该方法适用于马铃薯种 8 种必需氨基酸的测定。
Abstract:An ultra high performance liquid chromatography tandem mass spectrometry was established for the simultaneous determination of eight essential amino acids in Solanum tuberosum L.: valine, threonine, leucine, isoleucine, lysine, methionine, phenylalanine and tryptophan. The sample was extracted with ultrapure water and separated by Accucore HILIC column. The mobile phase was methanol solution containing 0.1%(volume fraction) formic acid, and the flow rate was 0.2 mL/min. Electrospray ionization source, positive ion scanning and multi reaction monitoring mode were used for detection. The eight amino acids had good linear relationships with chromatographic peak area in their mass concentration range, and the linear correlation coefficients were 0.999 0-0.999 6, the detection limits were 0.04-1.50 mg/kg and the quantitative limits were 0.12-4.99 mg/kg. The relative standard deviations of the determination results were 0.4%-4.6%(n=6), and the recoveries of samples at different addition levels were 89.9%-115.1%. The method is suitable for the determination of 8 essential amino acids in Solanum tuberosum L. seeds.
