猪肉冻干粉中克伦特罗标准物质的制备

Preparation for reference material of clenbuterol in pork freeze-dried powder

摘要:对活猪采用大腿肌肉注射法持续给予一定浓度的克伦特罗溶液,饲养至稳定状态后,宰杀取猪肉部分制备猪肉冻干粉中克伦特罗基质标准物质。样品检测参考GB/T 22286—2008《动物源性食品中多种β-受体激动剂残留量的测定》,采用同位素内标液相色谱质谱联用法检测克伦特罗的含量,并对样品中克伦特罗的均匀性和稳定性进行评价。定值采用同一检测方法8家实验室协作定值方式,并对标准物质的不确定度进行评估。结果表明,该标准物质的均匀性和稳定性满足标准物质制备的要求,标准物质中克伦特罗含量定值结果为(0.927±0.075)μg/kg(k=2,以复溶后肉糜计)。该标准物质可用于相关检测实验进行克伦特罗分析方法的确认和过程质量控制,并保证测试分析量值溯源的有效性和不间断性。
Abstract:Live pig was continuously given a certain concentration of clenbuterol solution by thigh intramuscular injection. After feeding to a stable state, the pig was slaughtered and the pork part was used to make the reference material of clenbuterol in pork freeze-dried powder. According to GB/T 22286-2008 Determination of Variousβ-agonists in Animal Food, the concentration of clenbuterol was determined by liquid chromatography mass spectrometry with isotope internal standard, and the homogeneity and stability of clenbuterol in the samples were evaluated. The specified value was determined by using the same detection method in 8 laboratories, and the uncertainty of the reference material was evaluated. The results show that the homogeneity and stability of the samples meet the requirements of reference materials, and the speci?ed value was (0. 927±0. 075)μg/kg (k=2), based on redissolved pork powder). This reference materials can be used in relevant detection experiments for the con?rmation of clenbuterol analysis method and process quality control, and ensure the e?ectiveness and continuity of traceability of analysis.
