摘要:以一起化学物质引起的食物中毒事件为例,建立超高效液相色谱-串联四极杆飞行时间质谱(UPLC-QTOF-MS)法快速筛查食物中有毒化合物的方法。用乙腈提取样品,采用基于QuEChERS原则的净化技术,以0.1%甲酸水溶液和0.1%甲酸-甲醇溶液为流动相进行梯度洗脱,经ACQUITY UPLC BEH C(100 mm×2.1 mm, 1.7 18 E +μm)色谱柱分离。采用全信息串联质谱(MS)模式对样品进行质谱(ESI)全信息采集,一次进样分析能够得到被分析物的一级和二级精确质量质谱数据,采用一级离子和二级质谱碎片进行标准质谱库匹配。在可疑食物及中毒人员血液中检出莫能菌素,确认了本次事件是由莫能菌素引起的食物中毒,建立了莫能菌素的质谱数据库。该方法适用于突发性化学中毒事件的快速筛查和检测。 
Abstract:A method of rapid screening of toxic compounds in food by ultra high performance liquid chromatography-tandem quadrupole time of flight mass spectrometry (UPLC-QTOF-MS) was established, with a case of a food poisoning incident caused by a chemical substance. The samples were extracted with acetonitrile, and the purification technology based on QuEChERS principle was adopted. The gradient elution was carried out with 0.1% formic acid aqueous solution and 0.1% formic acid-methanol solution as the mobile phase. The samples were purified by ACQUITY UPLC BEH C (100 mm×2.1 mm, 1.7μm) chromatographic column separation. The full information tandem mass 18 E + spectrometry (MS ) mode was used to collect the information of mass spectrometry (ESI ). The primary and secondary accurate mass spectrometry data of the analyte was obtained by one injection analysis, and the primary ions and secondary mass spectrometry fragments were used for standard mass spectrometry library matching. Monensin was detected in the blood of suspected food and poisoning personnel. It was confirmed that the event was food poisoning caused by monensin, and the mass spectrometry database of monensin was established. The method is suitable for the rapid screening and detectionl of sudden chemical poisoning events.
